Free ammo acids in senile cataractous lenses: Possible osmotic etiology

چکیده

Comparison of free amino acids in freshly excised rabbit lenses and in lenses taken from the contralateral eyes after storage for 24 to 48 hours reveals essentially no postmortem changes. This suggests that human eyebank lenses may be considered as normal with respect to the free amino acid pool. Analysis of senile cataractous lenses, compared to eyebank lenses, reveals a wide range of amino acid concentrations, from much higher to much lower than eyebank lens levels. Amino acid concentrations in cataracts decrease with decreasing lens weight, visual acuity, and per cent protein. Free proteogenic amino acid concentrations decrease from levels well above those in eyebank lenses, while nonproteogenie amino acids decrease only from normal levels. Thus selective accumulation of free proteogenic amino acids seems to be an early event in the etiology of senile cataract. This conclusion suggests the following hypothetical mechanism of senile cataractogenesis. Inhibition of protein synthesis, ivith continued normal proteolysis, could account for selective increase in proteogenic amino acid levels. Increased normal hydrolysis or pathological autolysis are possible but not essential explanations. Increased intracellular osmotic pressure and resultant i?nbibition of water may cause the wellknown early intumescence of senile cataract. If damage to cell membranes results, lens protein may be lost by leakage rather than by autolysis. Consequent dilution of cytoplasm and disruption of cellular structure are sufficient explanation of opacification.

Studies on protein and taurine in normal, senile and diabetic cataractous human lenses.

An attempt has been made to explore the possible role of Taurine in cataractogenesis. Normal lenses were obtained from eye bank donors and cataractous lenses from patients who had undergone surgery for cataract extraction. Lenses were weighed and homogenised. Extraction, isolation and estimation of protein and taurine were carried out. It has been found that the lens wet weight increased progre...

Lens superoxide dismutase and catalase activities in diabetic cataract.

OBJECTIVE Biochemical evidence suggests that the oxidative damage of the lens proteins is involved in the genesis of senile cataract and the development of diabetes-related pathologic changes such as the formation of cataracts. In particular, lens proteins are subject to extensive oxidative modification. Oxidative damage either decreases the antioxidant capacity or decreased antioxidant capacit...

Role of lipid peroxidation in the pathogenesis of myopic and senile cataract.

AIMS/BACKGROUND Increased production of free radicals, consumption of antioxidant, and oxidation of unsaturated lipids have been observed recently in cataractous lenses and active participation of the retina in human cataractogenesis has been proposed. To verify this hypothesis, the total (GSH) and oxidised (GSSG) glutathione concentrations were assayed in the lens and the malondialdehyde (MDA)...

Free epsilon amino groups and 5-hydroxymethylfurfural contents in clear and cataractous human lenses.

The free epsilon-amino groups and 5-hydroxymethylfurfural (5-HMF) contents were determined in soluble and insoluble proteins of clear human lenses and diabetic and nondiabetic senile cataractous lenses. The free epsilon-amino group content of soluble proteins in diabetic cataracts was decreased by 37% (P less than 0.01), whereas in nondiabetic senile cataracts it did not differ from that of cle...
